ENHANCED OSTEOGENIC DIFFERENTIATION OF HUMAN MESENCHYMAL STEM CELLS BY FLEXIBLE β-TCP/PLA BONE GRAFTS WITH SILICATE ADDITIVE

In recent years, ceramics, polymers, and composites have been used to develop biologically mechanically suitable bone scaffolds. β-tricalcium phosphate(β-TCP) is a widely bioceramic in tissue engineering. It shows excellent osteoconductivity, osteoinductivity, good biocompatibility properties, as its chemical composition similar the original structure of bone. Herein, we designed β-TCP-PLA composite scaffolds containing two different concentrations silicate additives. We aimed investigate effect silicate-additive with varying (0.8% 1%) on osteogenic differentiation human marrow-derived mesenchymal stem cells (hMSCs) seeded flexible grafts. The morphological β-TCP-PLA-based grafts was assessed by scanning electron microscopy (SEM) tensile strength evaluated. results showed that had porous structures. hMSCs evaluated alkaline phosphatase (ALP) activity DNA content measurements. Compared grafts, these synthetic 0.8% 1% significantly promoted proliferation differentiation. Moreover, increased ALP activity. outcomes present study suggest might be useful for encouraging regeneration
